Hey Andy thanks for your question. There are many possible variations that could exist in your question. If by "main panel" you mean the service equipment and the generator is for a standby system then a tap ahead of the service disconnecting means is allowed but that circuit must have its own service equipment as stated in 230.82(5). The service equipment overcurrent device location is stated in 230.91 so yes overcurrent protection is required.
